Jiadifenolide is an architecturally complex seco-prezizaane-type sesquiterpenoid isolated from the pericarps of Illicium jiadifengpi. It features a highly oxygenated cage-like architecture with seven contiguous stereocenters. The biological activity studies show that Jiadifenolide displays potent neurotrophic activity at very low concentration and is regarded as a promising lead compound for developing drugs for neurodegenerative diseases. The combination of a challenging cage-like motif and intriguing biological properties has promoted the development of efficient strategies toward their chemical syntheses. This report focuses on the recent advances in the total synthesis of Jiadifenolide.
